Hi all,
I am experiencing a very strong background on my IHC slides even on the control (where only the secondary antibody was applied). To block the non-specific sites, I used a sheep serum (3%) in PBS-T for three hours. However, it does not seem to work as there is still this background. I am thinking to try TCT or BSA as blocking solution. If anyone else has another idea of how to reduce the background, please, let me know! Thanks a lot for your help!
Nathalie Marquet Hi, try incubating the samples with sodium borohydride (NaBH4). This will help reduce the formaldehyde induced green fluorescent background.

Hello Nathalie, Are you sure that your slides never become dry during the procedure ... this also causes high fluorescent background. Keep them constantly wet. Best
