We are studying cell proliferation in a cell type (from a marine invertebrate) that is non-adherent. Does anyone have suggestions for method(s) to evaluate EDU labeling in non adherent cells? Fixing the cells to slides has not been working well.
I assume that poly-lysine slides did not work for you...
Then, you could try embedding the cells in low melting point agarose.
After fixing and washing, mix 1:1 with agarose (around 1%) and place in a mold or frame. I make thin square layers and then cut them smaller to make the sample more accessible. Proceed as a whole mount staining in eppendorfs or multiwell plates -more reagents are needed this way-. Maybe you need to increase some steps in the protocol, like washings.
Some people use gelatin instead or agarose. Good luck!
