I am working in stress physiology, since corticosterone is the primary biomarker for the stress response. I want to measure cortiocosterone levels in the pregnant rats. Anesthetizing rat during pregnancy, will it be adverse situation to the developing fetus? will it affect the healthy developing fetus in anyway?
if so, Which would be the suitable anesthesia for this study?

My question is that i want to collect blood from pregnant rat, for blood collection need to anesthetize rat, so i want to know that anesthetizing rat during gestation will affect normal development of fetus.
for aforesaid method leukocyte count also need blood collection.

You may sample the blood from the saphenous vein, which can be done without using anesthesia:

However, the sampling itself may be stressful (at least to some extend), therefore keep in mind to be relatively quick (sampling within ~3 minutes after taking the rat out of the cage), or otherwise your corticosterone levels might be elevated due to the stress response of the sampling itself.

Adrenal steroids are quite stable if bacterial conversion is prevented by adding thimerosal or NaAzide to the funnel of a metabolism cage. Measurement of corticosterone in 24h urine collections is easy by ELISA and provides an integrated sample of cort production throughout the day without restraint or pain. We found that a 48 h acclimatization period was usually enough to provide a steady baseline.

I would suggest measuring CORT in the fecal pellets!
