These day, I need to use IF to analyse VE-cadherin and Zo-1 expression to confirm the integrity of HUVECs layer.
However, it cannot work.
Can anyone help me solve it. I heard that we need some chemical, protein or creating stress to induce ZO-1 expression.? But I dont know what exactly it is?
I recommend you try the ZO-1 antibody from Proteintech, cat #: 21773-1-AP. I've successfully used this antibody in westerns and in fixed CLARITY perfused sections (by IF) and seen good results. This antibody is human specific, with some slight mouse cross-reactivity. You may want to make sure that what you have are really HUVECS as endothelial cells should express ZO-1. Of course all the normal caveats apply as well: make sure you're following all steps to prep your tissue/cells appropriately, the antibody is rated for the application and tissue type you're using it for, etc. Good luck.
