I am about to measure the secretion of a protein into the medium in a cell culture experiment (via exocytosis or extracellular vesicles). However, we have a certain level of death (10-20%) in our culture. Now the problem is how to discriminate between proteins that actively secreted and proteins that passively released due to death or death induced shedding. Normal cytotoxicity assays show the death rate, but do not give any clue about the impact of death on shedding and protein release. Is there any experience using a marker or an intracellular protein that we could employ as an indicator? My cells are neuron.
