I have used Lorke's method of determining LD50 of my test drug. The lowest dose according to this method is 10mg/kg and my mice died at this dose within the 30 minutes of IP injection. Does that mean my drug is too much toxic? and how should I calculate its ld50 value? The paper that im following for Lorke's method is also attached in this query
Determining LD50 of a drug is an obsolete concept, the data is not useful and there are publications on that. Instead MTD (Maximal Tolerated Dose) is calculated for drugs in acute toxicity studies, on the basis of which doses for repeat dose toxicity studies are determined.
You are using a very old reference, which is based mostly on oral route of administration. As you are administering drug through ip route, you have to move to lower doses as the drug is causing mortality at 10 mg/kg.
As I said earlier, LD50 is determined for chemicals and not for drugs. However, if you still want to continue, then use protocol mentioned in OECD guidelines 423/425, which are available free on internet.
