I am performing MTT assays to charactherize tumor cells resistance to chemotherapy (I am comparing the treated cells vs. control (non treated) from different tumor cell lines).
I have determined the IC50 for both groups, but I was wondering how do I know that the shift in IC50 is significant? For instance, for one linneage, the IC50control= 39.5µM and the IC50treated=268.4µM (almost 7x higher than control, which seems significant for me), while in another cell line, the IC50control=194.7µM and IC50treated=208.6µM (not so different...).
Do you perform any statistical test to determine if the shift in IC50 is significant?

I suspect that you are not differentiating between "statistical significance" and "practical importance".
Whether there is a meaningful difference in the means you mention, only you can determine if these are meaningful differences, given your field, the specifics of the situation being studied, and so on.
It's possible for two means to be statistically different, but be close enough to one another that no one would care from a practical point of view.
However, if you are interested in statistical significance, there are methods to assess this.
It is the concentration of the extract at which 50% free radicals are stabilized.
