Is anyone aware of a specific protocol in order to culture cancer cells from pleural fluid? I want to culture malignant cells from patients with pleural effusion.I have used Percol but with not good results. Please let me know if you have a protocol or specific instructions.Thanks
Its already in fluid and might not need dissociation or Percol. I would suggest to centrifuge the fluid, save supernatant, and resuspend the pellet in complete medium and try culture. It has to be done as soon as the fluid is collected. There could be very few cells in the beginning.
I agree with Goodwin. However be aware that the rate of establishment of the cell culture this way is very low, and in my experience there is not a lot that can be done to encourage growth.
We have used RPMI with 15% FCS, also DMEM in place of RPMI. Keep cells in T25 flasks (even after splitting) to encourage growth.
Tumour cell content in pleural effusion is highly variable, and you will have a lot of infiltrate.
Thanks for your replies. The reason why I used Percol was to seperate cancer from benign cells so they could grow faster on the plates. It is a slow process and I wondered if anyone had suggestions how to accelerate the growth.
