we have ctll-2 cell line. after defrizzing the cells, the third day on, the cells are dying for unknown reasons (with no color change in the medium). Could you give me a suggestion about this problem? we changed the medium, FBS and IL-2, but the problem is not resolved. Is it okay to add pen-strep antibiotic? and does mercaptoethanol need to be added to the cell culture medium?
Hi Arezoo, CTLL2 need stimulation to stay alive. ATCC recommends culturing the cells in media with T-STIM (with ConA). https://www.atcc.org/products/all/TIB-214.aspx#culturemethod

I totally agree with Nicole, it is essential to use t.stim within your medium. CTLL2 cells growing dependent upon the use of IL2.
Actually i am agrwoing ctll2 cells and they are totally grwoing well with good cell viability. and for their culture you should do the following; To make the complete growth medium, add the following components to the base medium: additional 2 mM L-glutamine; additional 1mM sodium pyruvate; adjust to a final concentration of 10% fetal bovine serum and 10% T-STIM with Con A.
