I am trying to do hydrolysis of DCFH-DA with 0.01 M NaOH. Stock is dissolved in anhydrous DMSO. Both the solvent and base is degassed.

The problem is dye seems to get hydrolyzed and oxidized at this step. solution is fluorescent within 5 mins. I tried using dry DMF and it didn’t help either.

I have been following steps described in so many articles. It’s Juz doesn’t seem to work for me! Has anyone worked with hydrolysis of the dye without cells?

We want to try to quantify ROS generated by our compound without cells. We can’t use lot of other compound because of overlap in absorbance and/or fluorescence.

Any advice is greatly appreciated!
