I am working on MCSF-7 and would like to know how to count cell with floe cytometry?How to gate and correctly interput and diffrenriate between life and dead cells? ( without using annixin)

On the other hand Live/dead discrimination methods are largely broken into 2 categories.
DNA binding fluorescent compounds such as Propidium Iodide and 7-AAD which bind DNA that is accessible in dead cells but not in live ones.
Amine Reactive dyes which bind both live and dead cells on the plasma membrane but can also bind proteins in the cytoplasm of dead cells making them far brighter. Many of these dyes are fixable and can have more options for channels than the DNA binding ones.
Annexin V will stain both dead and cells undergoing apoptosis.
