I have the absorbance values of an enzyme reaction taken over for 1 hour (for different substrate concentrations). Can anyone tell me how to calculate the velocity of the reaction to draw a michaelis-menton plot.

From the cell path length, molar absorptivity of the product and the rate of change of absorbance you can calculate the reaction rate,

Hi there,
Also make sure the reaction occurs linearly versus time over the period considered. Beer's law is applicable to the calculation of rate: (deltaOD/delta t)=epsilon.l.(delta concentration/delta t).

Thanks for answering the question. I used the microplate reader for the study. So as the path length, shall I take the depth of the well.
