1. how many exosomes I have? if I dissolved the total exosome pellet in 200ul PBS and I took 10ul from it and added it into 990ul filtered dh20 for nanosight analysis and the NTA analyzed this as 6.8x10e8/ml.
2. Now I want to make a co-culture of these exosomes with my selected cell lines.
sample 1 has yielded 6.8x10e8/ml exosomes (from NTA analysis)
sample 2 has yielded 8.8x10e8/ml exosomes (from NTA analysis)
I want to add the same amount of both the samples to my cell line how would I calculate?
For 1; multiplied the NTA value with the dilution factor; e.g. 10 microL to 1000 microL, the dilution factor is 100, prior to that 200 to 10 microL, so here the dilution factor is 20. Therefore the amount of exosome is = 6.8x10e8 x 100 x 20
For 2, dilute sample 2 with your media to 6.8x10e8
Hi Shahid, is the aim to add exactly the same number of exosomes to your target cells?
Simple way to do it is
[Target cell number] / [Exosome concentration] x [Desired number exosomes per cell] = Volume of exosomes in ml
So, for example, if you want to treat 1 x 10e6 cells with 100 exosomes per cell you would calculate
Prep 1: 1x10e6 / 6.8x10e8 x 100 = 0.147ml
Prep 2: 1x10e6 / 8.8x10e8 x 100 = 0.114ml
