How to adjust plate reader to read extinction 2.5 at 450 nm ?

I am using ELISA Kit to measure PYY in mouse serum by a colorimetric method. The instruction require Photometer for microtiter plate (Plate reader), which can read extinction 2.5 at 450 nm. My plate reader can be adjusted at 450 nm but I do not know what is the meaning of extinction 2.5 and how can I know if I can use my reader as it is or I have to adjust something in it and how ? My reader is SoftMax Pro 6.4 from Molecular Devices
