I am studying the secretion of the gut hormone (GLP-1) using the NCI-H716 cell line. Since it is my first time to use this cell line in particuljsr and suspension cell line generally, I am wondering how is usually treatments are added to cells in suspension. Is it just add the tested compounds to the media and centrifuge the cells out to collect the media after the incubation time? Or should we find a way to force the cells to adhere to the dish or the plate before adding our treatments?

You had better induce NCI-H716 cells to differentiate on matrigel-coated plate and they will attach on the bottom. You can find some references.
Yes, I do agree with kim if you want to get continuous secretion out of it. But then also there is few possibilities of getting cells in the spent media. So even after growing cells on matrix you should centrifuge the media in order to get rid of cells.
