Hi all,

I want to use phytagel for fluorescent microscopy imaging. Normally, I use agarose and sandwich it between two coverglasses until it solidifies then I place my sample (bacterial cells) on it.

I have prepared 1% phytagel with water and it did not solidify. I tried 1% phytagel with 1X PBS and it solidify when i put droplets on a surface however it did not solidify when I tried to put it between two coverglasses.

Anyone knows the trick?

Not: I used sigma protocol to prepare. After dissolving the phytagel in RT, I did microwave to melt it.
