If you need to have an idea of the traffic along a roas section or on a particular route, you'll probably have to use a traffic assignment model. For some countries / cities, statistics may also be available, based on collected cellular signals (used for instance in "live" GPS car systems.
If you do not have enough data from cellphones or a demand model you can count the traffic volume. This method is used to validate the models and you could count in the peak hours, all day, typical and atypical days.
Why make it so complicate? Use an automatic traffic recorder (ATR) (those road tubes) placed across the road for a week. Or how about just going out there with a clicker for a hour or two, and count passing vehicles.
