Has anyone looked into this aspect where you can replace target cancerous cells from the co- culture but at the same time use any stimulus to activate CAR-T cells in a cell-based IFNg assay ?
Hi Roychowdhury, I think you must use CD3/CD28 beads to activate and expand CAR-T cells without target and/or feeder cells in the culture, ok? All the best!! ;)
In addition to Mateus Souza-Barros's response, CD3/CD28 beads can be used to activate and expand CAR-T cells in a non-target-specific manner. However, if you are looking to observe antigen-specific activity, you may consider using a soluble antigen. For instance, if you have anti-CD19 CAR-T cells, adding sCD19 to the cells should result in activation and the release of cytokines.
