THP-1 cell is a suspension cell, so as I know we do not assign passage numbers. I want to know that for how long time we can keep culturing THP-1 cells after thawing from cell stock.
THP-1 are suspension, so they don't usually follow normal passaging rules. Keep them less than a million/mL, change medium regularly (2-3 times per week) and change out the culture every 2-4 months. Roughly 15-20 "passages." You know they've gone bad when they are no longer round and are showing small protrusions under a phase scope from the culture flask.
I agree with most of Jordan's response, though I would toss the cells after 2 months. The cells can be a bit heterogenous. Over time you'll see a population of larger cells expand. The cells may also begin to form grape cluster like colonies. Good luck!
Thank you Jordan and Michael for your comments. I have been using THP-1 cells since few months. Last week, the THP-1 cells did not respond to PMA well. I think, I cultured for too long time (more than 2 months). I started culturing from a fresh stock. I hope it goes well.
