After getting them out of the freezer (-20) and melting them in the fridge how long can the buffers (which contain skimmed milk) be stored there before use?
I have used 4 percent BSA for blocking which is stable for around a week in 1 X PBS at 4 degree, after which growth appears making the medium turbid. So u can check it before use.
preferably making fresh blocking buffer containing skim milk at the day you are doing your experiment. However, bb containing BSA 4% could stay up to 5 days stable in the fridge.
Freshly prepared Skimmed milk in TBS can be kept 3 days in the fridge, generally it takes seconds to make from powder using a vortex, so I suggest you always start from fresh. Alternatively if your primary antibody is compatible with NaN3 you can add some to your blocking buffer and keep in the fridge for weeks.
