I would like to be able to store single use aliquots of both for use in cell culture, dissolved in DMSO. I would dilute them with media/buffer on the day of use.
There is insufficient information on how stable the chemicals will be in solution, according to several manufacturers.
Has anyone used either in cell culture? If so, did you have to replace them very frequently? Have you noticed a difference in potency over periods of say, a few months?

I do not have direct experience with dexamethason and methylprednisolone as aliquotted stocks in DMSO. But with a lot of other less stable chemicals it proved to be possible to store them for prolonged periods (years) when dissolved in waterfree DMSO and stored aliquotted at minus 20C. I have used this scheme with some chemicals susceptible to oxidation (ROS indicators) with good results. Kind regards, Harrie Verhoeven.

I have experience only with dexa. In the lab we use 100%ethanol to dissolve dexamethasone, It works perfectly and we keep our aliquots for several months. I use this routinely in cell culture with no differences in potency (check GR target genes).

Up to 5-6 year time period is preferable, Still, some studies suggest preservation of stem cells up to 22.5yrs in 10% DMSO with FBS. The longer you store is inversely proportional to reviving rate.
