However, I am still concerned does a temperature needs to be maintained when the leaves are preserved in alcohol or they will be fine at room temperature?

Thanks for the valuable information. Yes currently I am extracting DNA from the samples preserved in 96% ethanol and it has been successful. The DNA quality is really good as mentioned in the research article shared by Mushtaq Ahmed.
