Hi all, I'm working with primary human synovial fibroblasts in culture. I have a couple of plasma membrane proteins which are upregulated nicely after 24 hours of treatment (quantified by immunofluorescence). I also want to stain for intracellular TNFa, but I'm aware that I need to use golgi block to prevent the TNF from being exported.
My questions are, how long can I treat the fibroblasts with golgi block without seeing an impact on viability? And can I treat with golgi block from the start? i.e. 24 hours before fixation.
