standard of surfactin is from sigma and it has 6 or 7 peaks in diagram, but there is just one or maybe 2 sharp peaks in my sample from Bacillus, can I decide and analysis my data based on these 2 peaks or should I have all 7 peaks and count based on that?

Dear Sir. Concerning your issue about the normal peaks in Surfactin and sample in HPLC . Previously reported method of identification and quantification of surfactin were time consuming and requires a large quantity of mobile phase. The new method was achieved by application of Chromolith® high performance RP-18 (100 × 4.6 mm, 5 µm) as the stationary phase and optimization of mobile phase ratio and flow rate. Mobile phase consisted of acetonitrile (ACN) and 3.8 mM trifluroacetic acid (TFA) solution of 80:20 ratio at flow rate of 2.2 mL/min was obtained as the optimal conditions. Total elution time of the obtained surfactin peaks was four times quicker than various methods previously reported in the literature. The method described here allowed for fine separation of surfactin in standard sample (98% purity) and surfactin in fermentation broth. Thanks Isam, exactly I used this method, but i mean that there is low similarity between surfactin peaks and samples peaks whereas my sample is Bacillus and it should produce the same peaks as standard.

I have a experience regarding the bio amine analysis , amino acid analysis and mycotoxin analysis with HPLC, LC-MS-MS and fatty acid profile analysis and pesticide analysis with GC, GC-MS. But I didn't have experience regarding the surfactin analysis. But in normally we injected the Standard sample/ reference sample/or IS to quantify our target compound. As a example, we used Amino acids premix-complex from Sigma and inject to the HPLC we found 19 primary and secondary amino acids. But we inject our sample to HPLC we couldn't found 19 amino acids. some time it may be 11 or 12. But we can quantify our results. In this case we inject individual amino acid to confirm the real amino acid found in our sample. I think that, you can count base on your 7 peak. But need to confirm it before analyzing.
