I have very little experience with histology. I have sections of spinal cord of my test groups (EAE mice) which are stained by the luxol fast blue-cresyl violet. what is the characteristic or appearance of the lesions? Which magnification is best? How I can best evaluate the results (quantitative)? Does anybody have a good paper or protocol with clear images and instructions?
A better option would be to stain for myelin for markers such as PLP. Adjacent slides can be stained for MHC or IBA-1 to visualize active microglia/inflammation.

I like to use solochrome cyanine as a myelin stain rather than LFB for qualitative images and use MBP antibody for quantitative analysis. They are both described in the paper above. Hope this helps!
It depends of the thickness of the sections. Luxol-Fast-Blue CV is a good staining. The myelated nerv fibers look like blue tubes. If there is any kind of demage you will see fragments, dots, some of them more intensiv stained then the other ones. Additionally I recommand a IHC with Anti Myelin-Basic-Protein (Anti-MBP, MAB382, Merckmillipore). Recommanded Magnifications: x20, x40, if it is nassyry x100 .
