Hi, I'm having problems growing the cell line PHM1-41 from myometrium. For it growth I'm using culture medium DMEM with its supplements (antibiotic and FBS). However they are growing very slow or they aren't. How do you usually grow it?
Your difficulty growing the PHM1-41 cell line from myometrium is perfectly understandable. Growing cell lines can be challenging, and it's great to see you're trying different approaches. Here's how I usually grow this cell line:
Subculture: Before growing the cells, I always subculture them and transfer them to a fresh culture medium. This helps maintain the health of the cells and ensures optimal growth.
Optimal culture conditions: I usually grow PHM1-41 cells in a humidified incubator at 37°C and 5% CO2. These conditions provide an optimal environment for cell growth.
Culture medium: For growing PHM1-41 cells, I use DMEM as the base medium, with antibiotics (such as penicillin/streptomycin) to prevent bacterial contamination and 10% FBS from providing the cells with essential growth factors.
Passaging cells: I usually pass them when they reach 70-80% confluency, which helps avoid cell death and maintain cell viability.
Monitoring cell growth: I monitor the cell growth regularly and adjust the culture conditions if necessary to ensure the best growth rate.
These are the steps that I follow to grow the PHM1-41 cell line. However, cell lines are known to be temperamental, and sometimes, you may need to try different cultural conditions to achieve optimal growth. If these steps don't work, you may need to try another culture medium or change the FBS source.
I hope this helps. Good luck with your experiments!
Now, we are using DMEM medium, supplement with 10 % FBS( with ambitiotic and antimicotic), but they have not been able to grow. We got it when they were in pass 13, from ATCC; however, right now they are in the pass 22, thus, they aren't very old.
