I know exact count possible by CFU/ml through spread plate count method & Haemocytometer count. But I want to know count bacteria & spore through spectrophotometer.But how is it possible give me suggestion.Otherwise i want to know how optical density result to convert spore & bacterial count.

yes its possible to count bacteria with spectrophotometer.
1. inoculate broth culture into fresh broth medium.
2. incubate it upto 24-48 hrs
3. after incubation take out the broth and take absorbance at 6oo nm with spectrophotometer (don't forget to keep uninoculated broth as control)
4. spread the inoculated culture (0.1 ml) on soild medium and keep the rest at 4 degree.
5. incubate the plates and after 24 hrs count cfu.
and thus you will get the ration 0.1 ml broth culture = xxx cfu/ml
