I obtained a curve and I will calculate IC50 from the formula ax + b but before the dosage I made a dilution of the extract 20 times
I wonder if I should divide the IC50 / 20 or not?
and how i can calculate the IC50 from ax + b?

Fitting the IC50 using the equation for a line (ax + b) is only suitable for a linear interpolation between 2 concentrations that are not very far apart. Otherwise, it is not a correct description of an IC50 curve, which has upper and lower bounds of 0 and 100% and should be fit by nonlinear regression to some variation on a Hill or logistic equation. For example: % inhibition = 100[X]n/(IC50n + [X]n) where [X] is the inhibitor concentration (not log-transformed) and n is the Hill coefficient.
The inhibitor concentrations should be those that were actually present in the assay when the reaction was occurring.

in a simple way, data the x-axis as concentration and the y-axis as the value of inhibition, then make a calibration curve y = ax + b. if y = 50, then Y is IC-50.

the dilution concentration used for the measurement of the inhibition percentage is the one used as the IC-50. no need to multiply or devide by 20
