I am performing an experiment to detect the presence of viral DNA in a cell line model. I am wondering when will be the best time to harvest the cells so as to achieve the highest viral load. Would it be 24 hours after passage? 48 hours? Do viral loads reduce or increase with each cell passage?
Any ideas or resources are welcome!

In general, if viral DNA is integrated within the cell genome, it is likely that the number would remain relatively stable for the time frame you indicated. If not, with increases in passage number, the load is predicted to decrease due to a number of factors. If you are more specific with respect to cell system and virus, it should be possible to address the question more specifically.
You should know the replication dynamics (biology) of your virus, and the properties of your cell line (some cells may be more susceptible to particular virus). About the time-frames, do you mean time after infection of the cells? Also, the viral load after 24h may depend on the dose of the infection of cells.
I agree with Faruk, it completely depends on the biology of your virus. For example if you are using a recombination deficient lentivirus then you can expect greatest load after 24 hrs then little further integration into the genome (and no replication). If your virus is WT and will replicate in the cells then the load should increase linearly until it reaches a plateau (no more cells left).
