I have attempted to crack my LbetaT2 cells twice but both attempts have failed. The second time, I know that I might have exposed the cells to trypsin for too long. However, I think there might be something else I am missing. Is there a specific kind of DMEM, PBS or trypsin I should be using? Currently I am using the high glucose Dulbecco’s Modified Eagle’s Medium

(DMEM) supplemented with 10% fetal calf serum (FCS) (Sigma-Aldrich, South Africa), 100 IU/mL

penicillin, and 100 μg/mL streptomycin, 1X PBS and 0.25% trypsin/0.1% EDTA.
