I was wondering how to detect the activation of the insulin pathway in a cell culture. I thought about quantifying the protein expression of phospho AKT or phospho GSK3 via Western Blotting, however there are a lot of protein isoforms and different phosphorilations... and I don't know which to choose.
Could you shed me some light about it?

Dear Juan
May I suggest to select a model in which insulin signaling pathway has been studied and use the same protocols than other scientists have used in such a model. In short, first the model and then looking for phospho AKT or GSK3 using the same tools that other scientists. Alternatively you may use different antibodies against different phosphorylated forms of those proteins (that are "correct") and check by yourself which are the good ones (I mean which are the appropriate phospho form(s) and the appropriate antibody(ies)).
