hi thanks for answer dear Dr.ahmad nadeem . but when cultivated neutrophil cells in the RPMI1640 isn't functional .do not match for test. such as DHR test.,
neutrophils are programmed for apoptosis and have only a few hours (5-7hrs) half-life in culture as Ahmed said. However, it might be possible to prolong their half-life by adding growth factors, I would try to add G-CSF if it is compatible with your experiment. Generally neutrophil activation prolongs ex vivo half-life.
See for example: Francois S et al. 2005 Inhibition of neutrophil apoptosis by TLR agonists in whole blood: involvement of the phosphoinositide 3-kinase/Akt and NF-kappaB signaling pathways, leading to increased levels of Mcl-1, A1, and phosphorylated Bad.
Another possibility might be to work with the neutrophil-like cell line HL-60 or PLB-985 for human. See this post: https://www.researchgate.net/post/Can_anyone_recommend_a_good_neutrophil-like_cell_line
