I am having difficulty trying to increase the concentration of both low molecular and high molecular weight Poly I:C without increasing the volume. Can anyone help?
Poly I:C is highly soluble in water and, from most good suppliers of synthetic poly I:C, most of it is detected as double-stranded. Therefore, one can dissolve the polymer in sterile water at a desired amount, keep the polymer at lower temperature (4 degree C), and read absorbance for nucleic acids (for single and double-stranded) to assess the concentration and the extent of double-strandedness.
