Good day everyone, Please how do I select a single clone, from a population of positive cells, I have done puromycin selection and now I want just a single clone from the cell population but I am having difficulties in doing this, please how can this be done. I will appreciate your kind response please. Thanks in anticipation !
We confirm cloning or select positive clones based on a few experiments, such as PCR using gene-specific primers or vector-specific primers, restriction digestion, and Sanger sequencing. Best wishes!
Are these eukaryotic or bacterial cells? For bacteria, you streak them on a plate at a low enough density for single colonies and then you pick a colony to grow up and test. All cells in a colony will be clones of one cell. For eukaryotes, we always used a cell sorter to isolate a single cell (mammalian). You use it to seed cells in a 96 well plate at a rate that will give you one cell per well and then you let them grow into colonies and test them.
