I was given HepG2 cell line to culture and even after changing media and leaving it to attach, none of the cells have attached. I added a picture of my culture. I was told that this cell line has been maintained with RPMI 1640 with 10% FBS even though I use DMEM for adherent cell lines but I followed what I was told and used RPMI but to no avail. What should I do?
We use DMEM supplemented with 10% FBS for HepG2 and it works perfectly. Maybe you can centrifuge and carefully resuspend the cells in a lower amount of DMEM and see if the attach. I would also definitely let them grow in higher concentration at least in the beginning.
I would recommend MEM with 10% FBS.
Anyway, make sure cells had been stored in apropriate conditions before you started working with them (maybe something went wrong during freezing procedure and because of that cells will never regain their properites?). Also, check whether your cell culture dishes are suitable.
