I had extracted prebiotics from plants through aqueous extraction process. Since to confirm the presence of prebiotics in plant samples I need molecular structure. Which technique will be best suitable for this? Thankyou.
Apart from the technics cited by Saeed Ayaz Khan, you can also use HPLC coupled to tandem MS/MS or UV/Vis but you should have an accurate method with a large database.
You can use LC/MS for volatile compounds and HPLC coupled to MS/MS or UV/Vis for non-volatile compounds. If you have pure compounds you can use NMR and FTIR.
First of all, the Thin Layer Chromatography should be used by spotting to identify the number of prebiotic compounds. Then, the mixture must be fractionated via column chromatography using different solvent systems. The suggested systems consist of ethyl acetate: ether in ratios ranged between 1:9 and 9:1. By applying the Thin Layer Chromatography, you can identify the presence of each compound in which fraction. Dry the detected fraction, measure its spectra using various available techniques.

In fact, the NMR, MS and ICP-AES techniques are more applicable for identifying the unknown chemical compounds and their structures. Among that, The UV-vis and XRD spectrometry are also prevalent. That depend to the objective and scale of the experiment.
Well you can use the GCMS and GCfID using a polar and non polar columns after the analysis the database from the computer will help you with the suggestions and you can calcul the retention time to confirm the compounds you have and you can help your self with the interpretation of the specter
