Suppose that I can mass culture an anaerobic or microaerophilic bacteria from mangrove sponge, are there any specific protocols for extraction using ethyl acetate in anoxic conditions? Or will the standard protocol of ethyl acetate in oxic conditions will do? I am more concerned on the death of the microorganisms after exposure to molecular oxygen considering the duration of the process which may affect the production and concentration of the secondary metabolites.
Hi Dan : This off course depends upon the scale of your work , but both can be controlled The Smaller Lab scale can be performed in a Glove box type of Hood system & the plant scale is fairly complicated but definitely , it will involve the costs as well . A well planned Nitrogen blanketing system must then be placed in the system to avoid the Aerobic oxidation of the anaerobic processes. The procedure has to be very well simulated , protected & controlled first by some rehearsals , it is not that simple.
