I need to calculate the pseudo first order kinetic parameters from the PL spectral data. The reactants include a fluorescent compound (A; used in excess) and different concentration of a fluorescence inhibitor (B). I have the data for F(max)-F(t) vs time. Can I use the formula ln(Fmax-Ft)=ln (Fmax)-kobs*t to calculate. I need to calculate k(obs) from the plot. Can anyone help me how to calculate the pseudo first order kinetic parameters from the data.
The best would be to fit your kinetic curves with F(max) and kobs as the variable parameters.A software for fitting are often built in the instrument software. If you send me the excel file, I might be able to help you.
At first to do such a task I recommend Matlab. Create a least square between the model output and experimental data. Then minimize that by manipulating the kinetic parameters. You can use Fmincon function or ga in Matlab.
Take a look to a program called DynaFit by a company called Biokin (it's free for academia). It's very powerful and relatively simple to use (waaaay easier than MatLab!)
