I need to measure glucose and lactose concentration quantitatively in defined media specifically. I have read about glucose measurement in HPLC but I only have C18 column, don't have refractometer. How can I possibly use HPLC or any method to measure glucose and lactose concentration?
Sigma Aldrich sells kits. Kit MAK017 uses glucose oxidase to measure glucose. Kit MAK097 uses an enzyme that converts lactose to galactose and glucose. Then an enzyme that oxidizes the galactose. Both make a color at A570nm. You can also measure glucose with hexokinase, to make glucose 6 phosphate followed by oxidation of glucose 6 phosphate and reduction of NADP to NADPH with glucose 6 phosphate dehydrogenase. There are probably kits for this too and maybe less expensive than the glucose oxidase kit. I don't know.
