Hi RG community, 

I am fairly new to cell culture to begin with but have been successfully working with primary human fibroblasts, but now I want to do experiments using primary human B-cells and crudely measure functional mitochondria with TMRE (for membrane potential) and mitochondrial mass (10-NAO) by flow cytometry, I would likely also image them with confocal. 

I have no background in immune cells or immune system biology really. I have tried to find information for the mitochondrial mass/content in B-cells but it has not been very fruitful. So in addition to information on how I would grow these cells in culture for at least 1 week if not more (i.e. what growth media, additives, and if they need stimulation what should I use), I would also like to know  whether or not B-cells contain a large number of mitochondria?

Please forgive my rambling and excessive questioning, I am still a rookie :)
