Does anyone know of an easily transfected cell line that is rich in mitochondria (amendable to mitochondrial study) and not immortal?
I want to generate cell lines expressing specific genetic variants in different genes and measure end points of mitochondrial function as well as senescence, if possible. I have only transfected HEK293 human cell lines before and I hear it is extremely difficult to do so in primary cells however transfecting immortalized cell lines and trying to study senescence is not feasible either.
If I understand your question correctly, you mean transfect a cell line (and not "transform"). By definition, a cell is immortalized if it acquires the ability to proliferate indefinitely. In this case, cells can be referred as a cell line (immortal). All cell lines can be transfected to varying extent and, depending upon the stimulus/signal and its strength can undergo senescence. For example, oncogene-induced permanent cell cycle arrest (senescence).
