I have been trying to culture cells from the tumour tissue sample I obtained. I homogenized the tissue sample and then seeded the sample into culture medium. But no result or there was a observable contamination. As well as I tried placing a tiny bits of excised tissue sample onto the culture flask and tried culturing them. But it's also no avail.
Your question is too general. What kind of tumour tissue did you obtain and how did you homogenize them?
Cell lines and primary cells are different things. Primary cells have a limited life span in culture and require specific nutrients to survive. What media are you using? What are you seeing them in? What cells are they?
You'd be better off culturing tumour-derived spheroids.
Jianqun Zheng I have homogenized them in 1xPBS/HBSS solution. Then did the following enzymatic and antibiotic treatment. Then tried culturing them in the culture media.
Ana Klisuric I am using DMEM-F12 medium. I use this media to usually culture SCC4 & SCC9 cell lines which requrires Hydrocortisone for better growth and survival. I followed the same post the tissue disaggregation. They are epithelial like cells of origin Squamous Cell Carcinoma
