Hello all,

I am working on genes affected in cholesterol metabolism in patients with Coronary Artery Disease and PCSK9 is one such target gene in my study. I have managed to obtain stable results with my housekeeping gene, 18S rRNA and the other target genes are also expressed moderately, with Ct values in the range of 18-29. Haven't had any major issues with those in both groups I'm studying. However, I am not able to optimise conditions for PCSK9, despite several attempts. I have used 100ng of diluted cDNA in both the experiments (Pictures attached) and did manage to get peaks, however, they were not consistent and the CV was affected between technical duplicates. Since there is ample literature stating that PCSK9 may have low expression in WBC's as compared to several other tissues, I tried to detect PCSK9 mRNA in endometrial tissue (which was available with another researcher in my lab), (Picture attached - Ct value had dropped to 28 in tissue from 31 in blood samples). Can anyone help me reach a conclusion wrt to PCSK9 expression in my samples?

I have used SYBr green chemistry for my assay.
