I would like to know apart from Raman and TEM analysis is there any other characterization techniques available to confirm the formation of graphene. Is is possible to grow graphene layers on a substrate at temperature below 900 degree C using CVD......

You can approximate No. of layers of graphene by evaluation from Raman Spectroscopy.
if the ratio of I2D/IG < 1 so it will suggest that we have few layers graphene (less than 5).
I2D/IG =1.2 so bi-layer Graphene
and I2D/IG > 2 single layer graphene.

Electric conductivity is the one of the important parameter to distinguish the nature of few layer graphen and mono/bi-layer graphene.
STM method is very useful - with this method for graphene single layer you will see 6 pi electrons in contrast to graphite where only 3 pi electrons are visible - because of ABAB stacking.
UV-vis spectroscopy also another option to find out number of layers of graphene. AFM also used for measuring thickness of the film and hence no. of layers. You can get available literature from internet. And see the following reference.....
