I'm extracting and quantifying anthocyanins in egg products and I would like clean up and get rid of all the phospolipids (basically phosphatidylcholine and phosphatidylethanolamine) before HPLC injection. Do you think that liquid extraction with chloroform will work?
I am not doing Mass Spec afterwards so maybe the presence of this phospholipids is not even a problem...
I would like to hear some opinions!

Hi Carlos, what ist your extraction solvent? If you use 100% methanol or acetonitrile you should try the phospholipidremoval well plates (or cartriges) from Phenomenes (phree), Waters (ostro) or Agilent (captiva). I use it for clean up eggsamples to determine coccidiostats. I had a lot of matrixeffects with the most of my analytes. I tested the 3 products, and now I use the phree and captiva. These both eliminate 98 - nearly 100% of the phospholipids. The faktor varies a liitle between the Lots / Charges, but it works very well.
Before pass through HPLC first conform the TLC with proper solvent systems.
After that continue for coloum chromatography for the separation of phospolipids.
Then characterize it by Mass spectra

Consider the use of a small strong anion exchange column or polyamide resin to capture the anthocyanins.

You are extracting anthocyanins. What form of anthocyanins are you looking for? Extraction efficiency depends a lot on their configuration, and the choice for the primaty solvent depends on this. So: what are you looking for?
