When we are infecting any gene ( For example A.tumefacines) into one cell line, the same cell line if i infect again and again up to diffrent transgenic lines, Can u please help me How to identify or check phenotypic diffrences in transgenic lines.

Hi Anil,
1. Be aware: when you do genetic transformation through tissue culture, remember that the process of 'tissue culturing' can also cause phenotypic abnormality. In other words, some of the phenotypic changes of T0 lines may not due to 'transgene' per se. Unless the change due to transgene is very obvious, such as the color change in the transgenic plants.
2. Some of the 'transgenes' might not cause any phenotypic and functional change any way; so if you try to identity 'transformants' from phenotypic change, you can end up nothing.

Hi Anil,
Yes, sure, if the transgene express (no gene silencing) and functional well. You will see that the transgenic lines develop disease resistant when you challenge/infect them with your microbes.

I attached this picture (from internet) for you. In the picture, left: non-transgenic, and on the 'right': transgenic lines. This just one example. You probably can see yours like these (depending on what damage your microbe can bring) if you really have a bona fide disease-resistant transgenic lines.
