I am giving an antibacterial treatment to bacteria and I am not sure how much time I have to treat bacteria for SEM analysis? and process for SEM fixation too
For SEM analysis, I apply the bacterial samples on the poly-L-lysine coated cover slips and fix them using 2.5% glutaraldehyde for at least 2 hours at room temperature.
for analysis SEM after action antiobiotic samples fixation 2,5% glutaraldehyd in the 0,1 M cacodylat buffer (pH-7,2), after 3-laving in the bufer postfixation 1% solution OsO4 in cacodylat bufer after dehydratation 30,50,70 ethanol application on the special metal-coat for prepartion in vacuumevoporator . After see samples in the SEM.
