I am interested in mechanism involving in seed germination, in that i want to quantify amino-acid , but i am not getting any colour with ninhydrin with methyl cellosolve with 55 % glycerol. is there anyone working in specific area?
Les acides aminés libres sont désaminés par la ninhydrine en libérant de l’ammoniac qui réagit avec la ninhydrine pour former un composé coloré (Moore et Stein, 1954).
A 500 µl d’extraits éthanoliques sont ajoutés 300 µl d’acide citrique 0.9 M, pH 5.2 et 600 µl d’une solution de ninhydrine 1.5 % (préparée à chaud dans un mélange éthanol 60 % / H2O, v/v, en présence de 40 mg d’acide ascorbique). Après agitation, incubation dans un bain-marie bouillant pendant 15 minutes et refroidissement rapide dans la glace, 2 ml d’éthanol 60 % sont ajoutés. La densité optique est ensuite mesurée à 570 nm. La glycine (Sigma) est utilisée comme standard.
