pT181 cop-634ts repC4 ori this is the ori site. I just wanna know whether it's compatible with probiotic organisms. How can I detect this, Kindly help me.
pT181 cop-634 repC4 has staphylococcous ori, an temperature senstive replicon.
This ori is used for replication in gram positive bacteria. If your interested probiotics organisms are gram negative in origin, then an appropriate vector to be used.
That's fine, my doubt is can I use the ori site of one organism to another. The organism which I'm working is also gram positive.Is there anything like ori site is specific to particular bacteria.
Most probably you might need a shuttle vector, if your are cloning a gene and then latter introducing into the probiotic bacteria. For gene cloning ,you need competent bacterial from primary from E.coli origin, hence shuttle vector.
I would think that the simplest way, assuming you have drug resistance on the plasmid, is to transform the bacteria of interest and test for drug resistant (and their relative frequency). If you have only few drug resistant, the wishful situation is that you may have mutant ori's compatible with your bacteria; the more likely explanation is that you have contamination...
