Macrophages and monocytes will be there when you isolate MSCs from cord blood, but as you continue your culturing only MSCs will proliferate and expand, whereas macrophages/monocytes are differentiated cells and they no longer proliferate or divide, and as u passage your cells only MSCs will be detached fast whereas most of the macrphages/monocytes will be retianed in the plate. Finally you can check the MSC lineage of your expanded cells by FACS or immunostaining with MSC surface markers.
I agree with Ms. Sivaraman's suggestion. Moreover, macrophages and monocytes firmly attached to the bottom of the plastic plates within 2-4 hours, and you can triturate the plate with culture medium to wash off MSC from the plate. Otherwise, continuous culturing and wait for macrophages and monocytes to die out within 2 weeks.
